E. coli grown on 15N salts for several generations produce labelled DNA that is more dense than normal DNA, as judged by density gradient equilibrium centrifugation. When transferred to 14N media, the first generation produces double-stranded DNA all of intermediate density, and the second generation DNA half of which is of intermediate density and half of normal density.
