A demonstration of semi-conservative DNA replication. E. coli grown on ¹⁵N salts for several generations produce labelled DNA that is more dense than normal DNA, as judged by density gradient equilibrium centrifugation. When transferred to ¹⁴N media, the first generation produces double-stranded DNA all of intermediate density, and the second generation DNA half of which is of intermediate density and half of normal density.
