Hanging-drop technique

A method for slowly changing concentrations in a protein solution in an attempt to prepare crystals. A small volume of protein solution (of the order of 5 μl) on a microscope coverslip is sealed drop-side down over a small well containing a larger volume (of the order of 1 ml) so that, over time, volatile solvents will exchange between the hanging drop and the solution in the well via the vapour phase and concentrate the protein solution or allow alcohol to pass over to it. The progress of crystallisation is observable in the droplet through the coverslip.
