Inverse PCR

A PCR protocol for amplification of sequences that flank a single known sequence. The source DNA is digested with a restriction endonuclease, circularised and ligated. Primers are constructed that will hybridise to the ends of the known sequence, but will be extended in opposite directions into the flanking sequences and around the circle. Cycles of polymerization, denaturation and rehybridization amplify the sequence between the restriction sites.
