Ligand-mediated PCR (LMPCR)

A method for amplification of a DNA fragment or a series of fragments with a single known primer-binding site. The fragment is denatured and hybridised with the primer. Extension of the primer with a polymerase that leaves a blunt end allows subsequent ligation to a linker-primer, which is a duplex oligodeoxynucleotide that will become the attachment site of a second primer in subsequent PCR cycles.
