Nested deletion sequencing
A method for extension of the length of a target DNA that can be sequenced using a single primer. Removal by an appropriate nuclease of varying polydeoxynucleotide lengths from the region near the primer binding site on a plasmid brings into sequencing range different regions of the target, followed by religation and circularization of the resultant shortened plasmids.
