TUNEL technique

The method of terminal deoxy-transferase (TdT) and dUTP assisted nick end labelling for detection of apoptosis in single cells. The method takes advantage of the fact that the DNA of apoptotic cells is extensively nicked. With TdT, biotinylated poly-U sequences are introduced at the nick sites and then conjugated with avidin bound to peroxidase. Peroxidase is used in an in situ chromogenic assay. The method is applicable to histological sections and to populations of cells, which can then be separated and analysed by flow cytometry.
