Cassette

A DNA fragment that contains a non-coding sequence (for gene knock-out) or a mutated replacement of a normal gene, and flanked by sequences homologous to those flanking the normal gene. By homologous recombination of its flanking sequences with the target genome, the cassette will replace the normal gene, albeit at a low frequency. The cassette may also contain genes that allow detection, e.g. the LacZ operon, and selection, e.g. neosporin resistance.
