An experiment designed to evaluate the contribution of different amino acids at a single sequence position to the stability of a polypeptide. Site-directed mutagenesis or synthetic chemical techniques are used to construct polypeptides that differ only at a single sequence position. Quantification of the stability of the polypeptide measures the contribution of each possible amino acid.
