A technique to select a bacterial strain with a desired genetic trait; a mixture of cells is streaked on to a solid support and the resultant colonies are screened (e.g. for antibiotic resistance or a DNA sequence complementary to a specific probe), and the selected colony is then restreaked for further rounds of purification.
