Transposon tagging

The use of a transposon to isolate a gene whose product may be unknown. Mutants in which a transposon inserts into and disrupts the function of a gene are selected by phenotype. A DNA fragment that contains the gene with its insertion is recognisable and is selected by its binding of a probe complementary to the transposon insert. The DNA whose sequence flanks the insert is isolated and used as a probe to screen a library of non-mutants for the intact gene.
