Phenol-emulsion reassociation technique (PERT)

A method to accelerate several thousand-fold the rate of DNA reassociation. An aqueous solution of single-stranded DNA with a chaotropic salt is shaken with phenol. When the resulting emulsion separates into two phases, the polynucleotide is found to have rapidly reassociated into double-stranded DNA.
