Primer extension analysis

A method for measurement of the amount of a specific mRNA and its length relative to a known sequence. A labelled synthetic primer designed for complementarity to a sequence in the mRNA, usually close to the 3'-end, is reverse transcribed to generate a cDNA that by the intensity of its labelling indicates the amount of mRNA originally present, and by its mobility on gel electrophoresis indicates its length, and hence the length of the mRNA from the site of primer attachment to its 5'-end.
